Comment fonctionne la SPME ?
Le principe de la SPME consiste à adsorber de façon sélective les composés volatils sur un polymère mis en contact avec l’espace de tête (le gaz) de l’échantillon. Suivant la nature de la fibre, le temps de contact, et la température de contact, l’adsorption des composés volatils peut être importante et nécessite souvent un travail d’optimisation. Il existe plusieurs type de polymère permettant d’adsorber les composés volatils, les plus connus sont le polydimethyl siloxane (PDMS) avec le divinyl benzene (DVB) et le carboxène (CAR). Certaines fibres ont même plusieurs polymère !
Une fois les COVs adsorbés sur la fibre, celle-ci est placé dans l’injecteur de la GC chauffé à 250°C généralement, pendant un temps donné, ce qui permet aux COVs ainsi piègés d’être relargués dans la colonne analytique.
Cette technique est extrèmement sensible et automoatisable, elle permet de détecter des composés volatils entre le ng/L et le µg/L.
L’analyse des terpènes et des COVs dans les plantes est une affaire d’experts.
La SPME une technique Polyvalente pour un screening large spectre
La SPME est tout à fait adaptée à l’analyse COVs dans des matrices solides ou liquides.
Dans notre laboratoire, nous utilisons la Solid Phase Micro Extraction (ou SPME) pour l’analyse d’extraits végétaux ou d’eaux florales car elle est à la fois sensible et polyvalente.
A TWISTAROMA nous disposons des outils permettant de réaliser des profils complets et larges spectres de ces produits grâce à notre passeur d’échantillons double bras équipé de cette technique développée par notre laboratoire.
Un protocole optimisé pour une répétabilité parfaite
Les profils SPME/GC/MS se font classiquement par la succession de l’extraction de l’échantillon via une fibre SPME puis la désorption de la fibre dans l’injecteur de la GC/MS qui nous sert alors à séparer et identifier les composés volatils préalablement extraits.
Les protocoles que l’on trouve dans la littérature préconisent généralement une dilution de l’échantillon par de l’eau salée avant son extraction par SPME, de plus la SPME nécessite l’ajout de témoin d’extraction pour normaliser les signaux et faire de l’analyse semi-quantitative.
Grâce aux passeurs d’échantillons il est possible de réaliser des séquences de plus en plus longues, jusqu’à 32 échantillons à la suite ! Avec une telle capacité il arrive que l’échantillon en position 32 patiente jusqu’à 32h avant d’être analysé ce qui amène de nombreux problèmes. En effet, durant notre R&D nous avons pu observer que les standards internes que nous ajoutions se dégradaient au fur et à mesure que les échantillons patientaient sur le portoir. De plus l’ajout de solution d’eau salée sur une matière première telle qu’un végétal peut conduire à l’oxydation de certains composés voire même à une extraction non souhaitée. En d’autre terme l’échantillon 1 n’a pas le même historique que l’échantillon 32. Grâce à nos équipements, il est possible de réaliser cette séquence « just in time », juste avant leurs extractions par SPME, on évite aux échantillons de rester sur le portoir. Une tête du passeur va alors ajouter juste avant l’extraction, la solution salée et le témoin d’extraction, puis l’autre tête équipée de fibre SPME va faire l’extraction. L’ensemble des échantillons est gardé à 4°C en attendant leur analyse.
Un exemple d'analyse par SPME : le houblon
Notre projet sur les analyses de plus de 150 variétés de houblons et leur bières associées nous ont conduit à mettre en place ce protocole en routine et nous sert dorénavant pour toutes les matrices alimentaires et cosmétiques à screener ou à analyser.
Le petit plus, cet outil nous sert aussi à cuire la viande (végétale ou non) juste avant son analyse grâce à nos incubateurs.
